چکیده

The 3D structures of all 12 mispairs formed in the active site of a DNA polymerase help explain their differential effects on polymerase stalling and on translocation of the primer terminus to the enzyme's proofreading site.

Transcriptional Pausing Caught in the Act

Single-molecule techniques now permit the tracking of a transcription complex along a DNA template in real time and to 1 bp resolution. As reported in this issue of Cell, Herbert et al. (2006) exploit this approach to study a central component of transcription regulation, the sequence-dependent pausing of RNA polymerase during transcript elongation.

Morbid Jealousy and Murder

tresses were unfaithful to them. As murderers go, they were an elderly group, of average age 474 years. Unlike ordinary, jealous lovers, their attacks were nearly always upon wives, rather than upon supposed rivals. The latter, being imaginary, could never be caught in the act and were only hazily identified. As with most deluded patients, these murderers seemed driven by emotional necessity to...

Crystal structures of open and closed forms of binary and ternary complexes of the large fragment of Thermus aquaticus DNA polymerase I: structural basis for nucleotide incorporation.

The crystal structures of two ternary complexes of the large fragment of Thermus aquaticus DNA polymerase I (Klentaq1) with a primer/template DNA and dideoxycytidine triphosphate, and that of a binary complex of the same enzyme with a primer/template DNA, were determined to a resolution of 2.3, 2.3 and 2.5 A, respectively. One ternary complex structure differs markedly from the two other struct...

How mobile are active RNA polymerases?

The photographs of 'genes in action' taken by Miller et al. (1970) are some of the most evocative in molecular biology. When bacteria are burst osmotically to spread their DNA, RNA polymerases can be seen frozen in the act of transcription as they processed along the template; the resulting nascent transcripts are covered with ribosomes, which are also caught in the synthetic act. These beautif...
